products are sold as \"human serum\". Plasma-derived serum starts from anticoagulated plasma that is later clotted artificially by adding calcium. The consequences are a different growth factor profile and residual citrate \u2014 which matters for MSC expansion, primary cells and calcium-dependent assays, and rarely matters for established cell lines.\n  <\/div>\n\n  <h2>How off-the-clot serum is made<\/h2>\n\n  <p class=\"lede\">No anticoagulant touches the blood at any stage.<\/p>\n\n  <ol class=\"steps\">\n    <li><strong>Whole blood donation<\/strong> into a plain tube or bag \u2014 no citrate, no EDTA, no heparin.<\/li>\n    <li><strong>Natural clotting.<\/strong> The coagulation cascade runs its physiological course. Thrombin is generated, fibrinogen converts to fibrin, and a clot forms.<\/li>\n    <li><strong>Platelet activation.<\/strong> As the clot forms, platelets degranulate and release their \u03b1-granule contents into the surrounding fluid \u2014 PDGF, TGF-\u03b2, VEGF, EGF and IGF-1 among them.<\/li>\n    <li><strong>Clot retraction<\/strong> under refrigeration, then centrifugation.<\/li>\n    <li><strong>Serum is drawn off<\/strong> the clot. What remains in solution is the serum, now depleted of fibrinogen and the consumed coagulation factors, and enriched with everything the platelets released.<\/li>\n  <\/ol>\n\n  <p>That last point is the one that matters commercially. <strong>The growth factors in off-the-clot serum are there because the clotting process put them there.<\/strong> They are not an additive.<\/p>\n\n  <h2>How plasma-derived serum is made<\/h2>\n\n  <p class=\"lede\">Clotting is prevented first, then induced later \u2014 out of sequence.<\/p>\n\n  <ol class=\"steps\">\n    <li><strong>Collection with anticoagulant<\/strong>, usually sodium citrate. Citrate works by chelating calcium, and calcium is required at several steps of the coagulation cascade.<\/li>\n    <li><strong>Plasma separation<\/strong> from the cellular fraction. Depending on the process, most platelets are removed at this stage.<\/li>\n    <li><strong>Recalcification.<\/strong> Calcium chloride is added to overcome the citrate block and force clotting.<\/li>\n    <li><strong>The fibrin clot is removed<\/strong>, and what remains is sold as serum.<\/li>\n  <\/ol>\n\n  <p>Chemically the product resembles serum. Three things are systematically different.<\/p>\n\n  <div class=\"split\">\n    <div class=\"box\">\n      <h3>Fewer platelet-derived growth factors<\/h3>\n      <p>If platelets were removed during plasma preparation, they were not present to degranulate when clotting was finally induced. The growth factors they would have released are simply absent.<\/p>\n      <p>This is the single largest functional difference, and it is why the two products can perform differently in the same culture.<\/p>\n    <\/div>\n    <div class=\"box\">\n      <h3>Residual citrate and altered ions<\/h3>\n      <p>Citrate is not removed \u2014 it stays in the product as calcium-citrate complexes. Sodium citrate also adds a sodium load, and recalcification adds chloride.<\/p>\n      <p>Free calcium in your medium is therefore lower than the nominal figure suggests.<\/p>\n    <\/div>\n  <\/div>\n\n  <h3>And a third, less discussed: residual fibrinogen<\/h3>\n  <p>If recalcification is incomplete, some fibrinogen survives the process. In practice this shows up as gel or fibre formation after freeze-thaw, or as stringy material in the culture vessel. Plasmapheresis \u2014 where plasma is collected and the cells returned to the donor \u2014 yields far more usable volume per donation and can be repeated much more frequently. A supplier who cannot answer it in writing is telling you something.<\/p>\n\n  <h2>One common confusion worth clearing up<\/h2>\n\n  <p><strong>Plasma-derived serum is not plasma.<\/strong> Plasma still contains fibrinogen and the full complement of coagulation factors. Availability drops with each additional constraint, so confirm current stock before you write the combination into a specification.<\/p>\n\n    <p class=\"fq\">How do I check what my current supplier delivers?<\/p>\n    <p>Look for \"off-the-clot\", \"from whole blood\" or \"no anticoagulant\" on the datasheet. If the wording is \"recalcified\", \"plasma-derived\" or \"from plasma\", it is not off-the-clot.
