Why BSEP testing matters in DMPK programs<\/h2>\n

\n The Bile Salt Export Pump (BSEP, ABCB11) is the primary hepatic\n transporter responsible for the efflux of bile acids from\n hepatocytes into bile. Inhibition of BSEP is one of the most\n established mechanisms underlying drug-induced cholestasis and\n is a recognised contributor to drug-induced liver injury (DILI).\n <\/p>\n

\n The vesicle-based assay uses taurocholic acid as a probe\n substrate to quantify BSEP-mediated transport into inside-out\n membrane vesicles, in the presence and absence of test\n compounds, allowing determination of inhibition potency and\n IC50 values.\n <\/p>\n

\n Because dual inhibition of BSEP together with MRP2, MRP3 or\n MRP4 can impair the compensatory bile acid efflux pathways that\n partially protect hepatocytes, a complete transporter panel\n alongside BSEP often provides a more complete DILI risk profile\n than BSEP testing in isolation.\n <\/p>\n <\/div>\n\n
