Spending extra on Low Endotoxin or Fatty Acid-Free grade for these applications adds cost without benefit.<\/p>\n<p>Standard BSA for cell culture supplementation at low concentrations (0.1\u20130.5%) as a media additive is also acceptable for most research applications where FBS is the primary supplement. When BSA is the sole protein supplement in serum-free medium, switch to Low Endotoxin grade.<\/p>\n<h2>Low Endotoxin BSA \u2014 serum-free medium and viral vector production<\/h2>\n<p>Endotoxin (LPS) in standard BSA activates TLR4 in mammalian cells \u2014 triggering NF-\u03baB \u2192 type I interferon \u2192 antiviral state. At the concentrations used in serum-free medium (1\u20135 g\/L BSA), even standard BSA with 5 EU\/mg endotoxin delivers 5,000\u201325,000 EU\/L to your cells \u2014 enough to chronically suppress AAV production, T cell expansion, and MSC proliferation.<\/p>\n<p>For serum-free medium where BSA is a primary component, specify Low Endotoxin (&lt;1 EU\/mg, LAL-tested per lot). This is not optional for AAV, lentiviral, and cell therapy applications where batch consistency matters.<\/p>\n<div class=\"qa\">\n<p class=\"q\">Is Low Endotoxin BSA the same as fatty acid-free BSA?<\/p>\n<p class=\"a\">No \u2014 these are independent specifications. Low endotoxin BSA controls LPS content but still contains normal levels of bound fatty acids (typically 0.3\u20130.8 equivalents of fatty acid per mole of BSA). Fatty acid-free BSA controls fatty acid content but may have normal endotoxin levels unless additionally specified as low endotoxin. For serum-free medium where both are critical, specify Low Endotoxin AND Fatty Acid-Free \u2014 some suppliers offer a combined grade.<\/p>\n<\/div>\n<h2>Fatty Acid-Free BSA \u2014 DMPK and lipid studies<\/h2>\n<p>BSA normally carries 0.1\u20130.8 mol of long-chain fatty acids per mol of protein, bound primarily at Sudlow sites I and II (the same sites that bind many drugs). For DMPK protein binding assays, fatty acid-free BSA is essential when:<\/p>\n<ul>\n<li>The drug competes with fatty acids for BSA binding \u2014 fatty acid-free BSA gives a higher unbound fraction (fu) that is more representative of true protein binding<\/li>\n<li>You are measuring fatty acid uptake or transport \u2014 endogenous fatty acids on standard BSA are background signal<\/li>\n<li>The assay is a fatty acid displacement assay \u2014 uses the fluorescent fatty acid analog ADIFAB; standard BSA gives false high displacement signals<\/li>\n<\/ul>\n<p>For DMPK vesicle assay buffers, fatty acid-free BSA is the standard \u2014 used at 0.5\u20131% to stabilise transporter membrane vesicles and reduce non-specific substrate binding without introducing competing lipids.<\/p>\n<h2>BSA pH 5.2 \u2014 a specialised grade<\/h2>\n<p>BSA adjusted to pH 5.2 is used in specific viral vector applications \u2014 particularly for stabilisation of AAV in low-pH formulation buffers and for certain cell transfection protocols where acidic pH improves endosomal escape.
