B. <a href=\"\/de\/fks-standard\/\">FBS Standard<\/a> oder <a href=\"\/de\/fks-premium\/\">FBS Premium<\/a>, bei 2\u20138 \u00b0C aufgetaut<\/li>\n<li>Aktivkohle, s\u00e4uregewaschen, f\u00fcr Zellkultur oder in Analysenqualit\u00e4t<\/li>\n<li>Dextran, mittlere Molmasse ca. 70 kDa (Dextran T-70)<\/li>\n<li>Puffer f\u00fcr die DCC-Suspension, z. B. 10 mM HEPES oder Tris-HCl pH 7,4 mit 0,25 M Saccharose und 1,5 mM MgCl\u2082<\/li>\n<li>Zentrifuge und f\u00fcr das Volumen geeignete R\u00f6hrchen oder Flaschen<\/li>\n<li>Wasserbad mit 56 \u00b0C (nur f\u00fcr die stringente Variante)<\/li>\n<li>Filter: 0,45-\u00b5m-Vorfilter und 0,2-\u00b5m-Sterilfilter, geringe Proteinbindung (PES)<\/li>\n<\/ul>\n<h2>Schritt-f\u00fcr-Schritt-Anleitung<\/h2>\n<h3>1. DCC-Suspension herstellen (am Vortag)<\/h3>\n<ol>\n<li>Eine Suspension aus <strong>0,25 % (w\/v) Aktivkohle und 0,025 % (w\/v) Dextran T-70<\/strong> in Puffer herstellen. Beispiel: 2,5 g Aktivkohle + 0,25 g Dextran in 1 l Puffer.<\/li>\n<li>\u00dcber Nacht bei 2\u20138 \u00b0C r\u00fchren, damit das Dextran die Aktivkohle beschichtet.<\/li>\n<\/ol>\n<h3>2. Aktivkohle pelletieren<\/h3>\n<ol start=\"3\">\n<li>Ein Volumen DCC-Suspension, <strong>das dem Volumen des zu behandelnden Serums entspricht,<\/strong> in ein Zentrifugenr\u00f6hrchen oder eine Flasche \u00fcberf\u00fchren.<\/li>\n<li>Zentrifugieren bei etwa <strong>500 \u00d7 g f\u00fcr 10 Minuten<\/strong>. Den \u00dcberstand vorsichtig abnehmen und verwerfen. Das Aktivkohlepellet bleibt im R\u00f6hrchen.<\/li>\n<\/ol>\n<h3>3. Erste Behandlungsrunde<\/h3>\n<ol start=\"5\">\n<li>Das aufgetaute Serum auf das Aktivkohlepellet geben und durch Vortexen oder Sch\u00fctteln vollst\u00e4ndig resuspendieren.<\/li>\n<li>Unter sanftem, kontinuierlichem Mischen inkubieren. Eine Variante w\u00e4hlen:\n<ul>\n<li><strong>Schonend:<\/strong> 12\u201316 Stunden (\u00fcber Nacht) bei 2\u20138 \u00b0C<\/li>\n<li><strong>Stringent:<\/strong> 45 Minuten bei 56 \u00b0C. Dabei wird das Serum zugleich hitzeinaktiviert (siehe <a href=\"\/de\/hitzeinaktivierung-serum-protokoll\/\">Protokoll zur Hitzeinaktivierung<\/a>).<\/li>\n<\/ul>\n<\/li>\n<li>Zentrifugieren bei etwa <strong>500\u20131.000 \u00d7 g f\u00fcr 10\u201315 Minuten<\/strong> und das Serum in ein neues R\u00f6hrchen mit einem frischen Aktivkohlepellet \u00fcberf\u00fchren (hergestellt wie in Schritt 2).<\/li>\n<\/ol>\n<h3>4. Zweite Behandlungsrunde<\/h3>\n<ol start=\"8\">\n<li>Erneut resuspendieren und mit derselben Variante wie in Runde 1 inkubieren. Eine zweite Runde senkt die Resthormonspiegel deutlich. \u201eDoppelt aktivkohlebehandeltes\u201c Serum wird zweimal behandelt.<\/li>\n<\/ol>\n<h3>5. Aktivkohle entfernen<\/h3>\n<ol start=\"9\">\n<li>Bei h\u00f6herer Drehzahl zentrifugieren, z. B. <strong>2.000\u20133.000 \u00d7 g f\u00fcr 20 Minuten<\/strong>, um die Aktivkohle m\u00f6glichst vollst\u00e4ndig zu entfernen.<\/li>\n<li>Das Serum \u00fcber 0,45 \u00b5m vorfiltrieren, dann \u00fcber 0,2 \u00b5m sterilfiltrieren. Feine Kohlepartikel verstopfen Filter schnell \u2013 planen Sie mehrere Vorfilter ein.<\/li>\n<\/ol>\n<h3>6. Aliquotieren und lagern<\/h3>\n<ol start=\"11\">\n<li>Das behandelte Serum aliquotieren und bei \u221220 \u00b0C lagern. Wiederholtes Einfrieren und Auftauen vermeiden. Jedes Labor legt seinen akzeptablen Restgehalt selbst fest.<\/li>\n<li>Wachstum testen: Behandeltes Serum unterst\u00fctzt das Zellwachstum schlechter als unbehandeltes.
