What is the difference between MCB and WCB?
Master Cell Bank (MCB): the original authenticated cell stock. MCB vials are used sparingly — only to create WCBs or in emergencies. Stored long-term with maximum security. Working Cell Bank (WCB): derived from the MCB at a defined passage number. Used for routine experiments and production runs. When WCB is depleted, thaw an MCB vial to create a new WCB. This two-tier system ensures original authenticated cells remain protected while providing practical stocks for daily use.
Why is lot reservation critical for cell banking?
Cell banking is a one-time event — once an MCB is frozen in a specific cryopreservation medium, changing to a new serum lot requires a new MCB preparation and full recharacterisation. Can rHSA replace FBS in GMP cryopreservation?
Yes — rHSA Premium Grade at 4–10% in defined cryobase medium provides xeno-free cryoprotection for GMP cell banking. The concentration must be optimised per cell type — 4% rHSA is typically sufficient for robust lines (CHO, HEK293), while iPSCs may require 8–10%. rHSA Premium Grade provides ISO 13485 documentation for GMP regulatory submissions. For clinical-grade iPSC banking, rHSA Premium + DMSO in DMEM/F12 is the standard xeno-free protocol.
Which serum for PBMC biobanking in clinical trials?
90% Human Serum AB Heat Inactivated + 10% DMSO is the standard for clinical-grade PBMC biobanking. FBS Very Low Endotoxin ≤1 EU/mL is acceptable for research-grade PBMC banking.
