Why MRP5 testing matters in DMPK programs
MRP5 (ABCC5) is a cyclic nucleotide efflux transporter with broad tissue expression, including cardiac and vascular smooth muscle tissue.
The vesicle-based assay uses cyclic GMP as a probe substrate to quantify MRP5-mediated transport into inside-out membrane vesicles, in the presence and absence of test compounds, allowing determination of inhibition potency and IC50 values.
MRP5 inhibition is assessed for compounds with structural similarity to cyclic nucleotide analogues, including certain antiviral and cardiovascular drug candidates. MRP5 testing is typically included in expanded transporter panels rather than the core regulatory set, depending on a candidate compound's structural class and target indication.
