Why MRP4 testing matters in DMPK programs
MRP4 (ABCC4) is a broadly expressed transporter with substrate specificity for cyclic nucleotides, bile acids and a range of nucleotide-analogue drugs.
The vesicle-based assay uses cyclic AMP or estrone sulfate as a probe substrate to quantify MRP4-mediated transport into inside-out membrane vesicles, in the presence and absence of test compounds, allowing determination of inhibition potency and IC50 values.
MRP4 inhibition is relevant both to hepatic bile acid efflux (alongside MRP3 as a compensatory pathway) and to the disposition of nucleotide-analogue antivirals and chemotherapeutics that are direct MRP4 substrates. MRP4 is one of several transporters assessed when a candidate compound is itself a nucleotide analogue, since MRP4 may mediate its own efflux as well as inhibit endogenous bile acid transport.
