Why MRP8 testing matters in DMPK programs
MRP8 (ABCC11) is expressed in a range of tissues including breast, liver and skin, with substrate specificity overlapping partially with other MRP-family transporters.
The vesicle-based assay uses cyclic GMP or estradiol-17β-glucuronide as a probe substrate to quantify MRP8-mediated transport into inside-out membrane vesicles, in the presence and absence of test compounds, allowing determination of inhibition potency and IC50 values.
MRP8 inhibition is assessed in specific drug efflux studies where structural or mechanistic overlap with other MRP-family substrates warrants a broader transporter screen. MRP8 is typically included in extended ABC transporter panels for compounds requiring a more comprehensive efflux risk characterisation beyond the core regulatory set.
