Description
Product Overview
SeamlessBio Single-Use Rocker Bags are film-based cultivation bags engineered specifically for rocking and wave-style bioreactor platforms (such as GE WAVE™, Sartorius BIOSTAT RM™, Thermo Fisher HyWave™, and Applikon). The gentle, low-shear rocking motion provides oxygen and CO₂ exchange without mechanical impeller agitation — the preferred cultivation format for shear-sensitive mammalian cell lines, microcarrier-based adherent cultures, stem cell expansion, and seed train workflows. Available in 10 L, 20 L, and 50 L working volumes, gamma irradiated to SAL 10⁻⁶, with fully customisable port configurations. Manufactured under ISO Class 7 cleanroom conditions with complete GMP documentation.
Available Formats
| Format | Working Volume | Primary Use |
|---|---|---|
| 10 L Rocker Bag | 10 L | Process development, clone selection, small-scale rocking bioreactor work |
| 20 L Rocker Bag | 20 L | Mid-scale seed train expansion and production workflows |
| 50 L Rocker Bag | 50 L | Scale-up campaigns and higher-volume rocking bioreactor production |
Why Rocking Bioreactor Bags vs. Stirred-Tank Systems?
| Parameter | Rocking Bag Bioreactor | Stirred-Tank Bioreactor |
|---|---|---|
| Agitation mechanism | Oscillating rocking motion — wave-induced mixing | Mechanical impeller rotation |
| Shear stress | Very low — suitable for shear-sensitive cells | Higher — can damage fragile cell types |
| Gas exchange | Surface aeration + headspace CO₂/O₂ exchange | Sparger-based — bubble-induced shear risk |
| Microcarrier culture | Excellent — gentle suspension of carrier beads | Possible but requires careful impeller selection |
| Cleaning & validation | None required — single use, gamma-irradiated | Full CIP/SIP cycles required |
| Scale-up flexibility | 10 L → 20 L → 50 L without platform change | Requires new vessel and re-validation |
Applications
| Application | Description |
|---|---|
| Shear-sensitive mammalian cell culture | CHO, HEK293, Vero, and BHK21 cell lines where impeller agitation causes unacceptable cell damage or productivity loss |
| Microcarrier-based adherent culture | Scale-up of adherent cells (e.g. Vero for vaccine production, MSCs) on Cytodex or similar microcarriers — rocking motion provides gentle bead suspension without excessive shear |
| Stem cell & primary cell expansion | iPSC, MSC, and primary cell types with high shear sensitivity — rocking platforms maintain viability and phenotype across passages |
| Vaccine production | Virus propagation on rocking bioreactor platforms using Vero or MDCK substrates — established manufacturing format for influenza, rabies, and viral vector vaccines |
| Seed train & process development | N-1 seed train steps, DOE screening, and clone selection at 10–50 L scale before transfer to stirred-tank production vessels |

Frequently Asked Questions
Which rocking bioreactor platforms are SeamlessBio rocker bags compatible with?
SeamlessBio rocker bags are engineered for compatibility with the major rocking/wave bioreactor platforms in use across the EU and globally, including GE Healthcare WAVE™ (now Cytiva), Sartorius BIOSTAT RM™, Thermo Fisher HyWave™, and Applikon rocking systems. The bag dimensions and port configurations are designed to match standard platform requirements. If you are using a less common platform, contact our team with the equipment model and required bag format — we can advise on compatibility or produce a custom-dimensioned bag.
Why are rocking bioreactor bags preferred for shear-sensitive cell lines?
Rocking bioreactor bags mix and oxygenate the culture by oscillating the bag on a rocking tray, creating a wave motion that distributes cells and nutrients without mechanical impeller agitation. This eliminates the high local shear zones that impeller tips create in stirred-tank systems, which can lyse fragile cells or trigger stress responses that reduce productivity. The format is particularly well-established for primary cells, iPSCs, MSCs, microcarrier-based adherent cultures, and virus production on sensitive substrates such as Vero cells.
Can rocker bags be used for microcarrier-based adherent cell culture?
Yes — this is one of the primary applications. The gentle rocking motion provides sufficient agitation to keep microcarrier beads (e.g. Cytodex 1, Cytodex 3, or similar) in suspension without the shear forces that cause bead-to-bead collision damage or cell detachment. Working volumes of 10–50 L are well-suited for scale-up from static flask or spinner flask culture to rocking bioreactor production. The 20 L and 50 L formats are frequently used for Vero-based virus production on microcarriers at clinical and commercial scale.
Are custom port configurations available?
Yes — all SeamlessBio rocker bags are available with customised port configurations. Standard configurations include inlet/outlet ports, a sparger tube for gas delivery, and a sampling port. Additional ports (vent filters, pH/DO sensor ports, harvest ports, Luer Lock sampling), alternative connector types (MPC, Luer Lock, AseptiQuik®), and custom tubing materials (TPE or platinum-cured silicone) can be specified on order. Contact our team with your process requirements and bioreactor platform model for a configuration recommendation.
What documentation is provided for GMP and regulatory compliance?
Every batch ships with a batch-specific Certificate of Analysis (CoA) and Certificate of Origin (CoO). A Leachables, Extractables & Toxicological (Tox) Validation Guide is available on request, along with USP Class VI compliance statements, ADCF declarations, and irradiation certificates (ISO 11137). All bags are manufactured in ISO Class 7 cleanroom conditions and gamma irradiated to SAL 10⁻⁶. Documentation packages support IND, CTA, and MAA submissions referencing single-use bag components.
