Exosomes · Extracellular Vesicles

FBS contains 1010–1012 EVs per mL. Every EV prep from serum-containing culture is co-isolated with bovine vesicles. If your downstream application requires MSC-specific EVs, the medium choice is not optional.

MSC-derived exosomes and extracellular vesicles are under active development as therapeutic delivery vehicles and biomarkers. The fundamental problem in most EV production workflows is serum contamination: bovine EVs from FBS co-sediment with MSC-derived EVs at every standard ultracentrifugation speed and cannot be removed without compromising yield.

CelliMax MSC1 is a serum-free, chemically defined MSC expansion medium that allows you to expand MSCs at sufficient density before switching to EV conditioning medium — or to run the full expansion in serum-free conditions from the start.

How CelliMax MSC1 fits into an EV production workflow

  • Expansion phase: expand MSCs in CelliMax MSC1 to target cell number — high proliferation rate maintained without serum
  • Serum-free switch: transition directly to EV-free conditioning medium with no serum withdrawal step — reduced stress response compared to serum-adapted cells
  • Scale-up: MSC1 supports growth in multi-layer flasks and can bridge to hollow-fibre bioreactor formats for larger EV volumes
  • Downstream clean-up: no bovine EV background — your NTA and cryo-EM data reflect your MSCs, not the medium

Documentation for EV therapeutic development

For groups developing MSC-EVs as an IND-track therapeutic or advanced diagnostic, full material documentation is available for CelliMax MSC1: composition disclosure, TSE/BSE declaration, endotoxin limits, sterility data. GMP-grade supply in powder and liquid formats.

Why bovine EVs are so hard to remove

Bovine serum EVs overlap with MSC-derived small EVs in size (roughly 30–150 nm), density and common tetraspanin markers. Differential ultracentrifugation, size-exclusion chromatography and tangential flow filtration all co-isolate them. “EV-depleted” FBS reduces but does not eliminate the background, and depletion also removes growth factors, so cells often grow more slowly. The cleanest approach is to keep serum out of the process altogether.

Practical notes for MSC-EV production

  • Harvest window: condition at 70–80% confluency; overgrown cultures release more apoptotic bodies and stress-derived vesicles.
  • Wash before conditioning: rinse the monolayer 2–3 times with PBS to remove residual medium proteins before adding conditioning medium.
  • Conditioning time: 24–72 h is typical; check viability at harvest (ideally above 90%) so the EV prep is not dominated by dying cells.
  • Pre-clearing: sequential low-speed spins (e.g. 300 × g, 2,000 × g, 10,000 × g) before ultracentrifugation, SEC or TFF.
  • Characterisation: report particle size and concentration (NTA), morphology (TEM/cryo-EM), positive markers (e.g. CD63, CD81, TSG101) and a negative marker such as calnexin, in line with MISEV recommendations.
  • Consistency: keep the expansion medium constant across batches — a defined medium makes EV yield and cargo comparable from run to run.

Frequently asked questions

Can I expand MSCs in FBS and only switch to serum-free medium for EV conditioning?

You can, but cells adapted to serum often respond to the switch with a stress phase that alters EV release and cargo, and serum proteins can carry over. Expanding in serum-free medium from the start avoids both effects.

Is EV-depleted FBS a sufficient alternative?

It lowers the bovine EV background but does not remove it completely, and the depletion step also changes the growth-factor content. For therapeutic or biomarker work, a defined serum-free medium is the more robust choice.
