Serum contains cytokines, growth factors and immunoactive proteins at undefined concentrations. If you are measuring IDO, PGE2 or TSG-6 from MSCs expanded in serum, you are measuring a mixture — not your cells.

MSC immunomodulation research depends on attributing paracrine signals to the cells, not the medium. Serum-based expansion introduces a background of bovine cytokines, complement fragments and immunoglobulins that co-purify with MSC conditioned medium and interfere with downstream immunosuppression assays.
CelliMax MSC1 expands MSCs in a chemically defined, protein-minimised formulation. The result is a defined baseline: what you detect in conditioned medium comes from your MSCs, not from serum carry-over.
Applications where defined expansion pays off
- Cytokine profiling: clean background for Luminex / ELISA panels — IDO, IL-6, IL-10, VEGF, HGF without bovine signal interference
- T-cell suppression assays: MSC conditioned medium from defined culture gives reproducible suppression curves — serum lots shift EC50 unpredictably
- PGE2 and COX-2 quantification: defined medium avoids arachidonic acid precursors from serum that drive background PGE2
- Licensing assays (IFN-g + TNF-a co-stimulation): consistent baseline MSC responsiveness across experiments — no inter-lot variability in starting immunophenotype
For conditioned medium (MSC-CM) production
If your workflow involves harvesting MSC-CM for downstream use — in co-culture, organoid or in vivo models — CelliMax MSC1 enables serum-free conditioning. The resulting CM has no animal-derived background. Volume pricing and GMP-grade supply available for larger CM production pipelines.
A clean licensing and readout workflow
MSC immunosuppression is largely inducible: inflammatory “licensing” switches on IDO, PGE2 and TSG-6. Defined expansion gives every experiment the same starting point.
- Licensing: IFN-γ alone or IFN-γ plus TNF-α for 24–48 h is the common approach; keep cytokine lots and concentrations fixed across experiments.
- IDO activity: measure kynurenine in the supernatant rather than relying on mRNA alone — activity is what drives T-cell suppression.
- PGE2 / COX-2: collect conditioned medium under serum-free conditions so serum-derived arachidonic acid does not inflate the baseline.
- T-cell suppression: CFSE- or dye-dilution proliferation of stimulated PBMCs or purified T cells at several MSC:responder ratios (for example 1:2 to 1:20) gives a usable dose–response curve.
- Assay medium: keep the co-culture medium identical between conditions — adding serum only at the co-culture step reintroduces the variable you removed during expansion.
Why serum distorts secretome data
Serum brings cytokines, growth factors, complement fragments and immunoglobulins that cross-react in ELISA or Luminex panels and co-purify with conditioned medium. Lot changes then shift EC50 values in suppression assays for reasons unrelated to the cells. Expanding and conditioning MSCs in a defined medium means the measured signal comes from the MSCs.
Frequently asked questions
Does serum-free expansion change MSC immunomodulatory capacity?
MSCs expanded in a defined medium keep their characteristic immunophenotype (CD73, CD90, CD105) and respond to IFN-γ/TNF-α licensing. Because the baseline is defined, differences between donors or conditions become easier to detect.
Can I use the conditioned medium (MSC-CM) directly in downstream models?
Yes. Conditioned medium from serum-free culture has no animal-derived background, which makes it suitable for co-culture, organoid and in vivo models.
Which readouts benefit most from a defined medium?
Cytokine panels (IDO, IL-6, IL-10, VEGF, HGF), PGE2 quantification and T-cell suppression assays — all readouts where serum proteins or lot effects otherwise add background.
Is a GMP grade available for clinical development?
Yes. CelliMax® MSC medium is produced under ISO 13485 at GMP grade, with full documentation for regulatory submissions.
