Description
Product Overview
SeamlessBio Chicken Serum — Normal Chicken Serum (NCS), EU Origin is collected from healthy Gallus gallus domesticus under controlled conditions, sterile filtered at 0.2 µm, and supplied with full lot documentation. It is the standard blocking and control serum for IgY antibody-based assays, where species-matched chicken matrix reduces non-specific background that mammalian sera cannot eliminate.
Normal Chicken Serum is used as a blocking reagent in IHC, IFA, and ELISA workflows using chicken IgY primary or secondary antibodies, as a species-specific cross-reactivity control in avian matrix assays, and as a supplement for insect cell culture and veterinary diagnostic assay development. See the full portfolio on our Animal Sera overview page.

Applications
| Application | Protocol | Note |
|---|---|---|
| IgY antibody blocking (IHC / IFA / ELISA) | 2–10% NCS in blocking buffer | Species-matched blocking serum for workflows using chicken IgY primary antibodies or anti-chicken IgY secondary antibodies — eliminates non-specific background that mammalian normal sera (goat, rabbit) cannot suppress in chicken IgY assays |
| Cross-reactivity and species-specificity control | NCS as avian matrix control | Negative control matrix for assays testing species cross-reactivity of antibodies or detection reagents — confirms absence of non-specific signal in avian serum background, relevant for multiplex assay development and anti-chicken secondary antibody validation |
| Veterinary diagnostic assay development | NCS as calibrator matrix or negative control | Species-matched matrix for calibrator and control preparation in poultry-specific ELISA, lateral flow, and immunoassay development — ensures matrix effects match field sample conditions in avian disease diagnostics |
| Insect cell culture supplement | NCS as serum supplement for Sf9 / Hi5 cells | Chicken serum is used as an alternative serum supplement for Sf9, Hi5, and other lepidopteran insect cell lines in baculovirus expression systems — relevant where FBS-based media show suboptimal performance or where mammalian-origin supplements must be avoided |

Frequently Asked Questions
What is chicken IgY, and why is Normal Chicken Serum used as a blocking reagent in IgY assays?
IgY is the predominant immunoglobulin class in chicken serum — the avian functional equivalent of mammalian IgG, but structurally distinct. Chicken IgY does not bind mammalian Fc receptors, which makes it highly useful in immunoassay formats applied to mammalian tissues where Fc receptor-mediated background is a problem with conventional rabbit or mouse IgG antibodies. When chicken IgY antibodies are used as primary detection reagents in IHC, IFA, or ELISA, the blocking serum must also be chicken-derived to saturate non-specific binding sites with species-matched protein. Mammalian normal sera (goat, rabbit, donkey) do not provide effective blocking for chicken IgY workflows — Normal Chicken Serum is the correct species-matched choice.
How is Normal Chicken Serum used as a cross-reactivity control?
In species-specificity testing, Normal Chicken Serum serves as the avian matrix negative control — confirming that a detection antibody or reagent does not produce non-specific signal against chicken serum proteins. This is relevant when validating anti-chicken secondary antibodies (confirming they react with chicken IgY but not mammalian IgG), when testing the species cross-reactivity of a primary antibody (does it recognise the chicken homologue of the target antigen?), and in multiplex assay development where the background signal of each species matrix must be independently characterised. The serum is pooled from non-immunised donors to provide a low-antibody, representative avian matrix baseline.
Can Normal Chicken Serum be used as a supplement for insect cell culture?
Yes — chicken serum is used as an alternative serum supplement for lepidopteran insect cell lines including Sf9 (Spodoptera frugiperda) and Hi5 (Trichoplusia ni) in baculovirus expression vector systems (BEVS). In applications where FBS-based media show suboptimal insect cell growth, or where mammalian-origin proteins in the culture supplement must be reduced, chicken serum provides an avian-origin alternative. It is most commonly used at 2–5% supplementation in Grace’s insect medium or TC-100. For standard industrial-scale baculovirus production, serum-free formulations are typically preferred, but NCS remains relevant for research-scale and optimisation workflows.
When is heat inactivation of Normal Chicken Serum required?
Heat inactivation (56 °C, 30 min) destroys complement proteins and is available on request. It is required when complement activation in the serum supplement would interfere with the assay or culture system — for example, in complement-sensitive cell lines, complement fixation test (CFT) controls, or ELISA protocols where complement-mediated lysis or background must be eliminated. For standard IgY blocking and cross-reactivity control applications, non-heat-inactivated NCS is typically used. Please specify the required form when requesting a quote.
