What does heat inactivation of FBS achieve?
Heating FBS at 56 °C for 30 minutes denatures and inactivates complement proteins (C1–C9 cascade). Complement can cause non-specific lysis of cultured cells, interfere with antibody-dependent assays, and affect virus neutralization tests. Heat inactivation removes this activity without significantly affecting albumin, transferrin, or most growth factors — making it the standard choice for immunological and virological applications.
Does heat inactivation affect FBS growth promotion?
Mild reduction in growth promotion is possible — heat treatment can partially denature some heat-labile growth factors. For most standard cell lines this is negligible. For highly growth-factor-dependent cells (e.g. primary cells, stem cells), non-heat-inactivated FBS is preferred unless complement inactivation is specifically required. We recommend testing both grades when switching protocols.
Which origin is best for heat inactivated FBS — US, Australia, or South America?
All three origins provide comparable growth promotion, protein content, and sterility for standard cell culture applications. Australian FBS has the lowest BSE risk (OIE-certified BSE-free country) and is preferred for vaccine production and the most stringent regulatory environments. US origin carries strong USDA documentation. South American FBS offers the most competitive pricing. Specify your preferred origin when requesting a quote.
Is heat inactivated FBS required for mycoplasma-sensitive applications?
Heat inactivation at 56 °C does not reliably eliminate mycoplasma — dedicated mycoplasma testing is required for that assurance. All SeamlessBio FBS lots are mycoplasma tested by PCR or culture method. Heat inactivation is specifically for complement removal; for mycoplasma-free assurance, rely on the mycoplasma test certificate rather than heat treatment.
