SeamlessBio FBS Tetracycline Free is fetal bovine serum specifically tested and confirmed free of tetracycline antibiotics (<detection limit by HPLC/ELISA). Available from South American, US, and Australian origin, each batch is sterile filtered at 0.1 µm with full lot-specific documentation including a tetracycline test report. It is the required serum supplement for Tet-On / Tet-Off inducible gene expression systems, where even trace tetracycline residues in standard FBS can suppress or trigger inducible promoters and invalidate experimental results. Why can’t standard FBS be used with Tet-On / Tet-Off systems?
Standard FBS frequently contains trace amounts of tetracycline antibiotics from livestock treatment. In Tet-On systems, doxycycline (a tetracycline derivative) activates gene expression — so residual tetracycline in standard FBS can cause leaky background expression even without intentional induction. In Tet-Off systems, the opposite problem occurs: trace tetracycline suppresses expression even when induction is intended. Tetracycline-free FBS eliminates this interference.
How is tetracycline absence verified in this FBS?
Each batch is tested for tetracycline and doxycycline by HPLC or competitive ELISA, with results reported below the assay detection limit. The test certificate is available with the CoA on request. This is the accepted standard for Tet-system validated FBS in academic and industrial laboratories.
Which origin should I choose for FBS Tetracycline Free — South America, US, or Australia?
All three origins are tested to the same tetracycline-free specification and provide equivalent performance for Tet-system applications. South American FBS is the most widely used and cost-effective choice. US origin is preferred where USDA documentation is required for regulatory submissions. Australian FBS carries the lowest BSE/TSE risk classification (OIE-certified, no BSE ever recorded) and is preferred for vaccine production, GMP bioproduction, and the most stringent regulatory environments.
