Description

Product Overview

SeamlessBio Mouse Serum — Normal Mouse Serum (NMS), EU Origin is collected from healthy Mus musculus under controlled SPF (Specific Pathogen Free) housing conditions, sterile filtered at 0.2 µm, and batch-tested for purity and consistency. Available as individual or pooled serum. Supplied with full lot documentation for use in immunoassay development, monoclonal antibody production, and murine cross-reactivity studies.

Normal Mouse Serum is the species-matched blocking reagent of choice when murine primary or secondary antibodies are used in ELISA, Western blot, and immunofluorescence assays. It is also widely used as a negative control matrix in antibody species-specificity testing and as a supplement in hybridoma cultivation for monoclonal antibody screening. See the full portfolio on our Animal Sera overview page.

Applications

ApplicationProtocolNote
ELISA blocking1–5% NMS in PBS or assay buffer before antibody incubationReduces non-specific background signal when murine primary or secondary antibodies are used in ELISA — species-matched blocking prevents cross-reactivity with endogenous mouse immunoglobulin binding sites
Western blot / IFA1–5% NMS in blocking buffer (TBST or PBST)Non-specific binding reduction in Western blot and immunofluorescence assays employing mouse-derived secondary antibodies
Hybridoma culture & mAb screeningNMS as culture supplement or assay matrixUsed in monoclonal antibody production workflows — NMS as a species-matched serum matrix in hybridoma screening ELISAs, or as supplement to support hybridoma cell growth
Cross-reactivity & species-specificity testingNMS as negative control matrixAntibody development and diagnostic kit validation — NMS confirms absence of cross-reactivity with murine proteins; reference matrix for assays targeting non-mouse analytes in a murine sample background

Frequently Asked Questions

What is Normal Mouse Serum (NMS) used for in immunoassay development?

Normal Mouse Serum (NMS) is primarily used as a species-matched blocking reagent in ELISA, Western blot, and immunofluorescence assays where murine primary or secondary antibodies are employed. At 1–5% concentration in PBS or TBS-Tween buffer, it saturates non-specific binding sites and endogenous mouse immunoglobulin binding sites before antibody incubation, reducing background signal. NMS is also used as a negative control matrix to confirm species specificity when developing or validating antibodies — it establishes the baseline signal in a murine serum background without target-specific immunoreactivity.

Is Normal Mouse Serum (NMS) the same as pre-immune mouse serum?

Normal Mouse Serum (NMS) is collected from non-immunised, healthy donor mice and contains only background levels of antibodies — no deliberate immunisation has taken place. It corresponds to what is described in the scientific literature as NMS or pre-immune mouse serum: a species-matched, antigen-naïve serum matrix. SeamlessBio NMS is sourced from SPF-housed donors, ensuring a defined and consistent baseline immunoglobulin profile suitable for use as a blocking reagent and negative control.

Is pooled mouse serum available, and what are the differences from individual serum?

Yes — both individual (single-donor) and pooled Normal Mouse Serum are available. Pooled serum is a blend from multiple donor animals, which averages out individual variation in immunoglobulin levels and protein composition, producing a more homogeneous and reproducible reagent. Pooled NMS is preferred for ELISA blocking and negative control applications where lot-to-lot consistency is important. Individual serum may be appropriate for specific applications where donor traceability is required. Please specify individual or pooled when requesting a quote.

Is heat-inactivated or gamma-irradiated mouse serum available?

Yes — both heat inactivation (56 °C, 30 min) and gamma irradiation are available on request. Heat inactivation destroys complement proteins and is recommended for complement-sensitive immunoassay applications and cell-based protocols where complement activation must be avoided. Gamma-irradiated NMS is used when additional sterility assurance is required. Both treatments are documented in the lot Certificate of Analysis. Please specify the required treatment when requesting a quote.

What documentation is supplied with EU Origin Normal Mouse Serum?

Every order includes a Certificate of Analysis (CoA), Certificate of Origin (CoO), and Safety Data Sheet (SDS). Additional documentation — sterility and mycoplasma test protocols — is available on request. Products are handled at ISO 9001 and ISO 13485 certified partner facilities. Batch reservation for up to 36 months is available on request to ensure lot-to-lot reproducibility for long-term assay workflows.
