What is the difference between Normal Lamb Serum (NLS) and adult Normal Sheep Serum (NSS)?

Normal Lamb Serum (NLS) is collected from Ovis aries up to 12 months of age. The key immunological difference is that lamb serum has lower endogenous IgG levels than adult sheep serum and may contain residual maternal antibodies transferred via colostrum during the first 6–8 weeks post-birth. For standard IHC and ELISA blocking with anti-sheep secondary antibodies, NLS and adult NSS are functionally equivalent — either can be used. NLS is specifically required when age-matched ovine serum is needed: neonatal immunology studies, colostrum immunity research, vaccine efficacy trials in lambs, and parasitology infection models where the age of the serum donor is scientifically relevant.

Can Normal Lamb Serum be used as a complement source in complement fixation tests (CFT)?

Yes — non-heat-inactivated NLS retains active ovine complement and is suitable as a complement source in complement fixation tests (CFT) and related haemolytic assays. When ordering for CFT applications, specify that heat inactivation is NOT required. Heat inactivation (56 °C, 30 min) destroys complement activity and is appropriate for blocking applications where complement activation must be avoided but is incompatible with CFT use.

Why is age-matched lamb serum required for vaccine efficacy studies?

In ovine vaccination trials conducted in juvenile animals, the pre-vaccination antibody baseline is age-dependent: lambs have lower immunoglobulin levels and a distinct antibody profile compared to adult sheep, partly due to the transition from maternally derived passive immunity to active immune competence. Using adult sheep serum (NSS) as the pre-vaccination baseline control in a lamb trial would introduce age-related matrix mismatch. Age-matched NLS — from animals of the same age range as trial subjects — provides the appropriate biological reference for valid titre comparison before and after vaccination.

Are age-defined pools of lamb serum available?

Yes — age-defined pools (for example, 0–4 weeks, 4–12 weeks, 3–6 months, 6–12 months) are available on request. Age-defined pools are relevant for neonatal immunology studies where the transition from colostrum-derived maternal immunity to active immune competence must be tracked across defined developmental stages.
