Why standard media fail in organoid culture
Standard cell culture media — DMEM, RPMI, MEM — are formulated for 2D monolayer growth on flat surfaces. In 3D organoid culture, cells exist in spatial gradients, form tight junctions, develop polarity, and require specific growth factor environments to differentiate correctly. Standard media fail because they do not provide the Wnt pathway support, EGF/Noggin/R-Spondin signalling context, or defined albumin carrier required for organoid self-organisation.
Standard FBS adds a second problem: lot-to-lot variability in undefined growth factor content, endotoxin levels, and hormonal components introduces unpredictable variability in organoid formation efficiency, size distribution, and differentiation state. In drug testing assays on patient-derived organoids (PDOs), this variability directly affects drug response measurements — making data non-reproducible between experiments.
OrganoMedium™ solves both problems: a chemically defined formulation specifically optimised for 3D organoid growth, certified under ISO 9001 and ISO 13485, produced in the EU, available in organ-specific variants (Brain, Intestinal, Liver) and a Xeno-Free variant for 3R-aligned and clinical translation work. Exclusively distributed by SeamlessBio and innoME in the DACH region.
For long-term monitoring: Conventional end-point assays on organoids miss the dynamics — growth rates, morphological transitions, stress responses, and death events that happen between sampling points are invisible. zenCELL owl captures these continuously, directly inside the incubator, across 24 parallel channels.

OrganoMedium™ GMP
ISO 13485-certified production. Full regulatory documentation package: COA, TSE/BSE statement, lot traceability. For clinical-grade and IND-relevant organoid workflows.
Serum Selection by Organoid Type
| Organoid Type | Application | Recommended Serum | Concentration & Note |
|---|---|---|---|
| Tumour spheroids (hanging drop / ULA) | Drug cytotoxicity, IC₅₀, cancer biology | FBS Low Endotoxin ≤5 EU/mL | 2–5% — reduced serum promotes spheroid compaction and hypoxic core formation |
| Tumour spheroids — antibody assays (ADCC) | Anti-tumour mAb screening on 3D models | FBS Ultra Low IgG <5 µg/mL | 2–5% — bovine IgG in standard FBS blocks Fc-receptor interactions on NK cells |
| Intestinal organoids | IBD, microbiome-gut interface, drug absorption | OrganoMedium™ Intestinal — serum-free | ENR defined conditions. rHSA or BSA Fatty Acid-Free as optional protein stabiliser. |
| Liver organoids / hepatospheres | DILI modelling, hepatotoxicity | OrganoMedium™ Liver or FBS Low Endotoxin ≤5 EU/mL | 2–5% for HepaRG spheroids. Serum-free preferred for primary human hepatocytes. |
| Brain / cerebral organoids | Neurological disease, neurotoxicity | OrganoMedium™ Brain — serum-free | Serum drives astrocyte differentiation — serum-free N2/B27 or OrganoMedium™ Brain required. |
| Pancreatic / colorectal PDOs | Personalised oncology drug testing | OrganoMedium™ Base — serum-free | Serum-free preferred — FBS undefined signals compromise PDO engraftment reproducibility. |
| iPSC-based organoids | Cardiac, neural, intestinal organoids from iPSC | FBS ES Cell Pre-Tested (early EB stage) → OrganoMedium™ | ES Cell Pre-Tested for EB formation; serum-free OrganoMedium™ for directed differentiation. |
| Cardiac spheroids | Cardiotoxicity testing | FBS Low Endotoxin ≤5 EU/mL — 2% maintenance | Low serum maintains cardiomyocyte contractility measurements — high serum masks force generation. |
zenCELL owl — Continuous Organoid Monitoring Inside the Incubator
Organoid cultures require continuous monitoring without disrupting growth. Removing organoids from the incubator for imaging — even briefly — causes temperature shock, CO₂ fluctuation, and mechanical disturbance that can alter growth trajectories and induce stress responses. zenCELL owl eliminates this problem: 24 channels of brightfield imaging directly inside the incubator, fully automated, across the complete culture period.
| Organoid Application | zenCELL owl Function | Key Advantage |
|---|---|---|
| Tumour spheroid growth | Automated size tracking, growth curves, morphology changes over days-weeks | No sampling required — continuous data from seeding to endpoint |
| Drug response testing | Time-resolved morphology before, during and after treatment | Identifies onset of response — time to 50% growth inhibition visible |
| Spheroid invasion / migration | Scratch assay compatible — automated migration zone analysis | Quantifies invasive capacity without fluorescence labelling |
| iPSC organoid differentiation | Morphology monitoring over weeks — structural changes during differentiation | Identifies failed differentiations early without destructive endpoint assay |
| HTS drug screening | 96/384-well compatible — full automation, compatible with Hamilton, Tecan, Beckman | True high-throughput organoid imaging without confocal infrastructure |
| Cardiac organoid contractility | Brightfield timelapse captures rhythmic contraction as visual quality indicator | Non-invasive functional assessment without calcium dyes |
| Long-term culture monitoring | Day 1 to week 8+ continuous — no gaps, no disruption | Complete growth history — reproducible culture protocols |
