Quick Definition

What is a 2D Bioprocessing Bag?: A 2D bioprocessing bag is a flat, pillow-style single-use bag made from multilayer polymer films, used for media preparation, buffer storage, and freeze-thaw of biopharmaceutical drug substance. The flat geometry provides uniform ice crystal formation during freezing, making 2D bags the standard format for controlled freeze-thaw of proteins, mAbs, and viral vectors.

Why 2D bags for freeze-thaw?

2D bags have consistent geometry enabling controlled-rate freezing systems (CRS) to apply a uniform temperature gradient across the entire bag. The flat format allows precise contact with CRS plates, ensuring reproducible freeze rates lot to lot. 3D cuboid bags perform poorly in freeze-thaw because variable thickness creates heterogeneous freezing rates.

2D vs 3D — when to use which

Use 2D bags for: freeze-thaw, media preparation <50 L, buffer storage, seed train. Use 3D bags for: large-volume buffer/media hold >100 L, bioreactor biocontainer. If freeze-thaw is required → always 2D.

Key Facts

  • Flat pillow-style construction from multilayer polyethylene-based films — typically 2–5 layers
  • Volume range: 150 mL – 50 L (SeamlessBio portfolio)
  • Primary applications: media preparation, buffer storage, drug substance freeze-thaw, seed train aliquoting, sampling
  • Flat geometry = uniform wall thickness = consistent ice crystal front during controlled-rate freezing → superior cryoprotection vs 3D bags
  • USP Class VI certified film materials, gamma sterilised (25 kGy), integrity tested per bag
  • Extractables/leachables data available on request for IND/BLA regulatory submissions
