Why Control testing matters in DMPK programs
The Control Vesicle Kit consists of non-transfected HEK293-derived membrane vesicles, processed identically to the transporter-expressing kits but without transporter overexpression.
The vesicle-based assay uses matched to the transporter kit under test as a probe substrate to quantify Control-mediated transport into inside-out membrane vesicles, in the presence and absence of test compounds, allowing determination of inhibition potency and IC50 values.
Control vesicles are essential for distinguishing transporter-specific uptake from passive diffusion or non-specific membrane binding, and are required for accurate IC50 and specificity calculations in any vesicle transport assay. We recommend running the Control Vesicle Kit alongside every transporter-specific kit in your panel to ensure background-corrected, specificity-validated results.
