NaivaCore is supplied as three separate components, each optimised for independent storage and flexible use. This architecture allows concentration adjustments, independent lot qualification, and stable long-term storage of thermolabile inhibitors.

Scientific Background
Naive vs. Primed Pluripotency
Human pluripotent stem cells exist in two distinct states: the primed state (corresponding to the post-implantation epiblast) and the naive ground state (corresponding to the pre-implantation inner cell mass). Naive hPSCs display key characteristics including dome-shaped colony morphology, X-chromosome reactivation in female lines, global DNA hypomethylation, and enhanced developmental potential.
Conventional culture media (E8, mTeSR, Essential 8) maintain cells in the primed state. Accessing the naive ground state requires active suppression of primed-associated signalling (MEK/ERK, PKC, WNT) while activating naive-supporting pathways (JAK/STAT3 via LIF, Nodal via Activin A).
The HENSM Approach
NaivaCore is based on the Human Extended Naive Stem Cell Medium (HENSM) formulation developed at the Weizmann Institute of Science (Bayerl et al., Cell Stem Cell, 2021). HENSM demonstrated robust naive reprogramming across multiple hESC and iPSC lines and introduced CGP77675 (SRC inhibitor) as a key addition enabling stable naive maintenance — an advance over earlier t2iLGö and PXGL formulations.

NaivaCore has been developed based on protocols validated across multiple hESC and iPSC lines. Compatibility across different genetic backgrounds should be confirmed by the user, as naive reprogramming efficiency can vary between lines.
Can I adjust the inhibitor concentrations in NaivaCore-SI?
Yes — this is a key advantage of the three-component format. Because NaivaCore-SI is supplied separately as a 100× concentrate, you can add it at varying dilutions to titrate individual pathway inhibition. This is not possible with single-formulation naive media.
What feeder system is recommended?
NaivaCore supports both feeder-free (Matrigel / Geltrex-coated plates) and MEF feeder-dependent culture. Geltrex Flex is included in NaivaCore-SI to support feeder-free naive colony attachment.
How does NaivaCore compare to t2iLGö or PXGL medium?
NaivaCore is based on the HENSM formulation which extends t2iLGö by adding SRC inhibition (CGP77675), which has been shown to improve naive maintenance stability. Unlike t2iLGö (which must be self-prepared from individual components), NaivaCore is available as a ready-to-use kit with consistent quality documentation.
