When is gamma irradiated FBS required?

Gamma irradiated FBS is required or strongly recommended for ATMP manufacturing (CAR-T, MSC, NK cells) where viral contamination of the final cell product poses patient safety risk; GMP manufacturing where the risk assessment demands validated viral inactivation steps; and clinical trial material production under IND where regulators expect additional adventitious agent controls beyond standard lot testing.

Does irradiation affect cell culture performance?

Gamma irradiation reduces growth factor activity by 10–30% depending on dose. Some cell lines show slower growth; others perform equivalently. Always validate irradiated FBS against your non-irradiated reference lot. Hybridoma cloning efficiency and HEK293T transfection efficiency are particularly sensitive to irradiation-related growth factor reduction.
